About Histamine

Histamine is a product of decomposition of histidine caused by the growth of certain bacteria in seafood. Fish containing high levels of histamine has been associated with many examples of poisoning commonly referred to as “scombroid poisoning,” a major health problem for consumers.

LDN´s trademark HistaSure™ stands for a group of assays based on ELISA or lateral flow technologies for the quantitative and/or semi-quantitative determination of Histamine in seafood – especially for all different kinds of fish samples. In addition LDN offers a colorimetric assay especially designed for fish meal samples: HisQuick™. The corresponding sample preparation protocols are based on simple extractions with water and follow the AOAC Official Method 937.07. On a regular base LDN participates at external quality assessment schemes such as CHEK and FAPAS – and to date always with success.

HistaSure™ ELISA High Sensitive (Cat#: FC E-3100)

This assay measures Histamine quantitatively in each kind of fish and fish products such as fresh fish (tuna, anchovy, mackerel, herring, salmon;), canned fish, salted fish, fish in oil, and fish meal.

Quality control measures designed to minimize the occurrence of scombrotoxic fish require the determination of histamine levels in the range of approximately 10 to 200 ppm. Good quality fish contain less than 10 ppm histamine, a level of 30 ppm indicates significant deterioration, and 50 ppm is considered to be evidence of definite decomposition. The defect action level (DAL), the level at which regulatory actions are taken for histamine, is 50 ppm (P. L. Rogers, W. F. Staruszkiewicz, Journal of Aquatic Food Product Technology, Vol. 9 (2) 2000 p. 5 - 17

In a comparative study where different commercial Histamine test kits had been compared the HistaSure™ ELISA High-Sensitive showed the best correlation to the HPLC-method and the authors concluded that this kit “is found to be a reliable method for testing histamine in TFPs (traditional food products) either for regulatory or HACCP applications in monitoring such hazard” [Sevim Köse et al., Commercial test kits and the determination of histamine in traditional (ethnic) fish products-evaluation against an EU accepted HPLC method; Food Chemistry Volume 125, Issue 4, 15 April 2011, Pages 1490–1497].

The LDN HistaSure ELISA Fast Track has been validated and certified as a Performance Tested Method SM (#021402 ) by the AOAC Research Institute as an effective method for the quantitative determination of histamine in fresh/frozen yellowfin tuna, canned tuna-chunk light in water, fresh/frozen mahimahi, canned sardines in oil and fish meal.

HistaSure™ Fish Rapid Test (Cat#: FC L-3300)

This assay kit is intended for the rapid screening of histamine in different kinds of samples, such as fresh fish (tuna, anchovy, mackerel, herring, salmon), canned fish, salted fish, fish in oil etc.

Histamine testing in fish is a possible control strategy that can be used by seafood processors in their HACCP program to address the hazard of scombrotoxin formation.

This kit combines the unique immunogold labeling technique with highly specific immunoreagents which results in a sensitive and flexible test system: Cut-offs can be adjusted quite easily depending on the requirements. HisQuick™ Histamine (Cat#: FC P-3000)

The HisQuick TM Histamine Rapid Test is a colorimetric assay for the quantitative determination of Histamine in fish meal.
The test kit contains enough reagents to determine 48 fish meal samples.
First, Histamine is extracted from the sample and then bound to an ion-exchanger column to separate it from impurities. Next, the Histamine is derivatized and the resulting dye is determined at 450 nm. The optical density of the dye is directly proportional to the concentration of the Histamine in the sample.
